Loss of motivation stems from disruption in the brain's dopaminergic reward pathways, particularly connections between the ventral tegmental area, nucleus accumbens, and prefrontal cortex. Dopamine, the neurotransmitter central to drive and pleasure anticipation, becomes depleted or less responsive due to chronic stress, depression, sleep deprivation, or lack of meaningful rewards. The anterior cingulate cortex, which helps assign value to goals and sustain effort, shows reduced activation.
